LA ACUICULTURA EUROPEA EN UNA PUJANTE Y MODERNA INDUSTRIA EN CONSTANTE EXPANSION. NO OBSTANTE, SE ESTIMA QUE LAS PERDIDAS ECONOMICAS DEBIDAS A ENFERMEDADES SUPONEN UN 20% DE LA PRODUCCION Y EN CONCRETO LOS PARASITOS ESTAN CADA VEZ MAS IMPLICADOS EN ELLAS. EL MONOGENEO SPARICOTYLE CHRYSOPHRII CONSTITUYE EL PRINCIPAL PROBLEMA PARASITOLOGICO DE LA ACUICULTURA ESPAÑOLA Y MEDITERRANEA, AUNQUE SU IMPACTO ECONOMICO ES DIFICIL DE CUANTIFICAR. ESTE PARASITO SE ANCLA EN LAS BRANQUIAS DE LA DORADA (SPARUS AURATA) Y SE ALIMENTA DE LA SANGRE DEL HOSPEDADOR PRODUCIENDO DAÑOS HISTOPATOLOGICOS, ANEMIA, EMACIACION, LETARGIA Y MORTALIDAD. LA MORTALIDAD APARECE CUANDO LA PREVALENCIA E INTENSIDAD DE INFECCION SON ALTAS, Y DEPENDE DE FACTORES BIOTICOS Y ABIOTICOS, COMO LA EDAD, LAS CONDICIONES DEL AGUA Y EL MANEJO DE LAS GRANJAS._x000D_ EL CONTROL DE LAS INFECCIONES Y EL DESARROLLO DE HERRAMIENTAS TERAPEUTICAS Y PROFILACTICAS ESTA LIMITADO POR GRAVES VACIOS DE CONOCIMIENTO SOBRE LA BIOLOGIA Y FISIOLOGIA DEL PARASITO Y LA INTERACCION PARASITO-HOSPEDADOR. EL PROYECTO SPARICONTROL PRETENDE CONTRIBUIR A LLENAR DICHOS VACIOS MEDIANTE ESTUDIOS IN SILICO, IN VITRO E IN VIVO SOBRE EL PARASITO Y SU RELACION CON EL HOSPEDADOR. EL OBJETIVO FINAL ES ENCONTRAR SOLUCIONES PARA MITIGAR EL IMPACTO DEVASTADOR DE LAS INFECCIONES POR S. CHRYSOPHRII EN LAS GRANJAS DE DORADA. LOS OBJETIVOS ESPECIFICOS SON: 1) ESTANDARIZAR UN MODELO DE INFECCION IN VIVO EN LA DORADA Y DE CULTIVO IN VITRO DEL PARASITO; 2) DESARROLLAR Y VALIDAR PROTOCOLOS FIABLES PARA EL DIAGNOSTICO DE LA INFECCION A GRAN ESCALA; 3) GENERAR METODOS DE DIAGNOSTICO PREDICTIVOS DE LA INFECCION Y LA CARGA PARASITARIA; 4) IDENTIFICAR LAS MOLECULAS DEL PARASITO IMPLICADAS EN SU PATOGENICIDAD, DEMOSTRAR SU ACTIVIDAD, E IDENTIFICAR SU PRESENCIA EN LA SANGRE DEL PEZ; 5) IDENTIFICAR, EVALUAR Y SELECCIONAR DIANAS TERAPEUTICAS Y CANDIDATOS TERAPEUTICOS; 6) REALIZAR PRUEBAS DE CONCEPTO CON LOS TRATAMIENTOS SELECCIONADOS EN INFECCIONES EXPERIMENTALES; 7) REALIZAR PRUEBAS DE CAMPO DE TRATAMIENTOS EN GRANJAS ENZOOTICAS; 8) DEFINIR LOS PRINCIPALES AGENTES IMPLICADOS EN LA INTERACCION PARASITO-HOSPEDADOR. EL PROYECTO TIENE UN ENFOQUE MULTIDISCIPLINAR, YA QUE INCLUYE TECNICAS DE CULTIVO IN VITRO, PARASITOLOGIA CUANTITATIVA, INTEGRACION DE DATOS MASIVOS, CITOQUIMICA, INMUNOHISTOQUIMICA, ZIMOGRAFIA, GENOMICA, TRANSCRIPTOMICA, PROTEOMICA, MICROSCOPIA ELECTRONICA Y CONFOCAL, ¿DRUG DISCOVERY¿, ENCAPSULACION EN ALIMENTOS, ENSAYOS DE TOXICIDAD, TRATAMIENTOS, ETC._x000D_ LA CONSECUCION DE ESTOS OBJETIVOS SUPONDRA AVANCES EN LOS CAMPOS DE LA INMUNOLOGIA DE PECES, PARTICULARMENTE SOBRE INMUNIDAD MUCOSAL DE LAS BRANQUIAS, MICROBIOMA DE LAS BRANQUIAS, ASPECTOS HASTA AHORA NO EXPLORADOS DE LOS MONOGENEOS, COMO SU PROTEOMICA O SUSCEPTIBILIDAD A DISTINTAS DROGAS. ADEMAS, SE APORTARA CONOCIMIENTO Y HERRAMIENTAS QUE PERMITIRAN MITIGAR EL IMPACTO DE ESTA INFECCION A CORTO PLAZO, COMO LA ESTANDARIZACION Y SIMPLIFICACION DE LOS METODOS DE DIAGNOSTICO, INCLUYENDO METODOS NO LETALES INDIRECTOS, O LOS METODOS ALTERNATIVOS DE TRATAMIENTOS. A LARGO PLAZO, LA IDENTIFICACION DE NUEVOS TRATAMIENTOS PERMITIRA SU DESARROLLO Y REGISTRO PARA USO COMERCIAL. ESTOS OBJETIVOS SON INNOVADORES Y NO SOLO APLICABLES AL MODELO PARASITO-HOSPEDADOR EN ESTUDIO, SINO QUE SERIAN UTILES PARA OTROS PECES INFECTADOS POR MONOGENEOS SIMILARES A NIVEL MUNDIAL. LA INTEGRACION DE TODA ESTA VARIADA INFORMACION CONTRIBUIRA A LA MEJORA DE LA SALUD Y BIENESTAR ANIMAL Y DE LA PRODUCCION ACUICOLA. (Spanish)

EUROPEAN AQUACULTURE IS A LARGE AND MODERN INDUSTRY WITH A HIGH INCREASING POTENTIAL. ECONOMIC LOSSES DUE TO INFECTIOUS DISEASES IN FISH FARMING ARE ESTIMATED AT 20% OF THE PRODUCTION VALUE AND PARASITES ARE INCREASINGLY RESPONSIBLE FOR THEM. THE MONOGENEAN SPARICOTYLE CHRYSOPHRII IS THE MAJOR PARASITOLOGICAL PROBLEM FOR MARINE SPANISH AND MEDITERRANEAN AQUACULTURE, ALTHOUGH ITS TRUE ECONOMIC IMPACT IS DIFFICULT TO QUANTIFY. THIS PARASITE ATTACHES TO THE GILLS AND FEEDS ON HOST BLOOD PRODUCING HISTOPATHOLOGICAL DAMAGE, ANAEMIA, EMACIATION, LETHARGY AND MORTALITY. MORTALITY APPEARS WHEN PREVALENCE AND INTENSITY OF INFECTION ARE HIGH AND IT IS DEPENDENT ON BIOTIC AND ABIOTIC FACTORS, SUCH AS AGE, WATER CONDITIONS, AND FARM MANAGEMENT._x000D_ MANAGEMENT OF THE INFECTIONS AND DEVELOPMENT OF PROPHYLACTIC AND THERAPEUTIC TOOLS ARE HAMPERED BY EXTENSIVE KNOWLEDGE GAPS ON THE PARASITE BIOLOGY, PHYSIOLOGY AND HOST-PARASITE RELATIONSHIPS. THE SPARICONTROL PROJECT INTENDS TO BREACH SOME OF THESE GAPS BY FOCUSING ON IN SILICO, IN VITRO AND IN VIVO CONTROLLED STUDIES ON PARASITE PHYSIOLOGY, ¿OMICS¿ AND HOST-PARASITE INTERACTIONS. THE FINAL AIM IS TO FIND SOLUTIONS FOR MITIGATING THE DEVASTATING IMPACT OF S. CHRYSOPHRII INFECTIONS IN MEDITERRANEAN FARMS. THE SPECIFIC OBJECTIVES ARE: 1) TO STANDARDIZE AN IN VIVO INFECTION MODEL IN GSB AND IN VITRO CULTURE OF THE PARASITE; 2) TO DEVELOP AND VALIDATE PROTOCOLS FOR RELIABLE AND HIGH-THROUGHPUT DIAGNOSIS OF THE DISEASE; 3) TO GENERATE NON-LETHAL DIAGNOSTIC PREDICTORS OF INFECTION AND PARASITE LOAD; 4) TO IDENTIFY PATHOGENESIS-ASSOCIATED MOLECULES IN THE PARASITE, TO DEMONSTRATE THEIR ACTIVITY, AND TO IDENTIFY THESE MOLECULES IN FISH BLOOD; 5) TO IDENTIFY, EVALUATE AND SELECT DRUG TARGETS AND THERAPEUTIC CANDIDATES; 6) TO CONDUCT PROOF OF CONCEPT TRIALS OF SELECTED TREATMENTS (DRUG AND PLANT-BASED) IN EXPERIMENTAL FACILITIES; 7) TO CONDUCT PROOF OF CONCEPT TRIALS OF SELECTED TREATMENTS (PLANT-BASED) IN FIELD STUDIES; 8) TO DEFINE THE MAIN HOST PLAYERS IN THE HOST-PARASITE INTERACTIONS. FOR THIS PURPOSE, A MULTIDISCIPLINARY APPROACH IS ENVISAGED INCLUDING IN VITRO CULTURE TECHNIQUES, QUANTITATIVE PARASITOLOGY, BIG-DATA INTEGRATION, CYTOCHEMISTRY, IMMUNOHISTOCHEMISTRY, ZYMOGRAPHY, GENOMICS, TRANSCRIPTOMICS, PROTEOMICS, TRANSMISSION ELECTRON AND CONFOCAL MICROSCOPY, DRUG DISCOVERY, FEED ENCAPSULATION, TOXICITY ASSAYS, INFEED TREATMENTS, ETC._x000D_ THE ACHIEVEMENT OF THESE OBJECTIVES WOULD ADD NEW INFORMATION ON FISH IMMUNOLOGY, PARASITOLOGY, MONOGENEAN PROTEOMICS, PARASITE DRUG SUSCEPTIBILITY, GILL MUCOSAL IMMUNITY AND MICROBIOMICS. IN ADDITION, IT WILL PROVIDE KNOWLEDGE AND TOOLS USABLE FOR MITIGATING THE IMPACT OF THE INFECTION IN THE SHORT TERM. THESE INCLUDE STANDARDIZED PROCEDURES FOR PARASITE DIAGNOSTICS AND PARASITE LOAD MEASUREMENT (INCLUDING NON-LETHAL INDIRECT METHODS), OR KNOWLEDGE ON THE EFFECTIVITY OF ALTERNATIVE TREATMENTS (BASED ON VEGETAL EXTRACTS AND DIET SUPPLEMENTS), NOT AFFECTED BY PHARMACOLOGICAL REGISTRATION PATHWAYS AND THUS AVAILABLE IN THE SHORT TERM. ON A LONGER TERM, THE IDENTIFICATION OF NEW TREATMENTS CAN START THE PATH FOR THE DEVELOPMENT AND REGISTRATION OF MUCH-NEEDED EFFECTIVE AND SAFE THERAPEUTICS. THE OBJECTIVES ARE INNOVATIVE AND NOT ONLY APPLICABLE FOR THE CONSIDERED HOST-PARASITE MODEL, BUT ALSO FOR OTHER FISH SPECIES AND SIMILAR MONOGENEAN INFECTIONS WORLDWIDE. THE INTEGRATION OF THE MULTIFACETED INFORMATION WILL CONTRIBUTE TO AN IMPROVEMENT OF FISH HEALTH, WELFARE, AND PRODUCTION. (English)
