DEBIDO A VARIOS FACTORES RELACIONADOS CON LAS RESTRICCIONES AL USO DE AGENTES QUIMIOTERAPICOS EN ACUICULTURA, A LA VIRULENCIA, Y AL CURSO EXTREMADAMENTE AGUDO DE LA ENFERMEDAD, HASTA EL MOMENTO NO SE DISPONEN DE MEDIDAS QUIMIOTERAPEUTICAS PARA SU CONTROL. LA UTILIZACION DE ALGUNAS MEDIDAS PREVENTIVAS DE HIGIENE Y DE INMUNOPROFILAXIS, SON LAS UNICAS HERRAMIENTAS DISPONIBLES ACTUALMENTE PARA CONTROLAR ESTA ENFERMEDAD. AUNQUE EXISTEN VACUNAS QUE UTILIZAN EL CILIADO COMPLETO EMULSIONADO EN ADYUVANTES OLEOSOS QUE HAN DEMOSTRADO UNA BUENA PROTECCION, SIN EMBARGO, ESTAS FORMULACIONES PUEDEN PROVOCAR UNA SERIE DE EFECTOS SECUNDARIOS ADVERSOS QUE HACEN QUE LIMITEN SIGNIFICATIVAMENTE LA PRODUCCION DE LOS PECES VACUNADOS. EN EL PRESENTE PROYECTO, PRETENDEMOS DESARROLLAR VACUNAS MENOS REACTOGENICAS QUE LAS CONVENCIONALES, COMPUESTAS DE ADUYANTES POLIMERICOS BIODEGRADABLES CARENTES DE TOXICIDAD Y DE UNA SELECCION DE ANTIGENOS DE MEMBRANA DEL PARASITO DE ELEVADA INMUNOGENICIDAD E INVOLUCRADOS EN LA PROTECCION PRODUCIDOS EN SISTEMAS DE EXPRESION EN EUCARIOTAS. PARA LA SELECCION DE LOS ANTIGENOS SE RECURRIRA A TECNICAS DE PROTEOMICA Y TRANSCRIPTOMICA, MEDIANTE LAS CUALES SE CARACTERIZARAN LAS PROTEINAS DEL PARASITO MAS RELEVANTES EN LA PROTECCION, SE PROCEDERA A LA SINTESIS QUIMICA DE LOS GENES CODIFICANTES DE DICHAS PROTEINAS Y, EVENTUALMENTE, SE MODIFICARAN AQUELLAS SECUENCIAS MEDIANTE MUTAGENESIS PUNTUAL PARA OBTENER TODAS LAS VARIANTES INTRAESPECIFICAS PRESENTES EN LAS DIFERENTES CEPAS Y CONSEGUIR UN ANTIGENO UNIVERSAL, QUE SE EXPRESARA EN SISTEMAS DE EXPRESION EN LEVADURAS Y MICROALGAS. LA VACUNA RECOMBINANTE CONTENDRA VARIAS PROTEINAS QUE SE EXPRESARAN EN TANDEM, EVALUANDOSE SU POTENCIAL ACTIVIDAD PARASITICIDA Y PROTECTORA MEDIANTE ENSAYO IN VITRO DE ACTIVACION DE LA COAGULACION/ACITIVACION DEL COMPLEMENTO Y MEDIANTE ENSAYOS IN VIVO TRAS LA INFECCION EXPERIMENTAL Y EVALUACION DE LA SUPERVIVENCIA. SCUTICOCILIATOSIS IS A SERIOUS DISEASE AFFECTING TURBOT AND OTHER FLATFISH IN CULTURE PRODUCED BY PHILASTERIDES DICENTRARCHI, A CILIATE ORIGINALLY FREE-LIVING THAT CAN BE TRANSFORMED, UNDER CERTAIN CONDITIONS, INTO A HISTIOPHAGE PARASITE CAUSING LARGE MORTALITIES IN INFECTED FISH. DUE TO SEVERAL FACTORS RELATED TO THE RESTRICTIONS ON THE USE OF CHEMOTHERAPEUTIC AGENTS IN AQUACULTURE, VIRULENCE, AND THE EXTREMELY ACUTE COURSE OF THE DISEASE, NO CHEMOTHERAPEUTIC MEASURES HAVE YET BEEN AVAILABLE FOR THEIR CONTROL. THE USE OF SOME PREVENTIVE HYGIENE AND IMMUNOPROPHYLAXIS MEASURES ARE THE ONLY TOOLS CURRENTLY AVAILABLE TO CONTROL THIS DISEASE.ALTHOUGH THERE ARE VACCINES THAT USE WHOLE CILIATES EMULSIFIED IN OILY ADJUVANTS THAT HAVE DEMONSTRATED GOOD PROTECTION, HOWEVER, THESE FORMULATIONS CAN CAUSE A SERIES OF ADVERSE SIDE EFFECTS THAT CAUSE THEM TO SIGNIFICANTLY LIMIT THE PRODUCTION OF THE VACCINATED FISH. IN THE PRESENT PROJECT, WE INTEND TO DEVELOP VACCINES LESS REACTOGENIC THAN CONVENTIONAL VACCINES, COMPOSED OF BIODEGRADABLE POLYMERIC ADJUVANTS LACKING TOXICITY AND A SELECTION OF PARASITE MEMBRANE ANTIGENS OF HIGH IMMUNOGENICITY AND INVOLVED IN THE PROTECTION PRODUCED IN EUKARYOTIC EXPRESSION SYSTEMS. FOR THE SELECTION OF THE ANTIGENS, PROTEOMIC AND TRANSCRIPTOMIC TECHNIQUES WILL BE USED TO CHARACTERIZE THE MOST RELEVANT PARASITE PROTEINS IN THE PROTECTION, THE CHEMICAL SYNTHESIS OF THE GENES CODING FOR THESE PROTEINS WILL BE CHARACTERIZED AND, EVENTUALLY, THOSE SEQUENCES BY POINT MUTAGENESIS TO OBTAIN ALL THE INTRASPECIFIC VARIANTS PRESENT IN THE DIFFERENT STRAINS AND TO OBTAIN A UNIVERSAL ANTIGEN, WHICH WILL BE EXPRESSED IN YEAST AND MICROALGAE EXPRESSION SYSTEMS. THE RECOMBINANT VACCINE WILL CONTAIN SEVERAL PROTEINS THAT WILL BE EXPRESSED IN TANDEM, EVALUATING THEIR POTENTIAL PARASITICIDAL AND PROTECTIVE ACTIVITY BY IN VITRO ASSAY OF COAGULATION / COMPLEMENT ACTIVATION AND BY IN VIVO ASSAYS AFTER EXPERIMENTAL INFECTION AND SURVIVAL EVALUATION.
